Preparation & Notes
"One Heparinized Plasma tube required. (Minimum plasma volume needed: 0.5 ml) Note: USE ONLY GLASS TUBE TO ALIQUOT. DO NOT USE PLASTIC TUBES. Follow the special procedure in collecting and processing the sample for PlHgb: Use a 20-gage needle to collect the sample. The patient must be relaxed. Place tourniquet on the upper arm. Puncture vein with as little trauma as possible. Draw one green heparinized top tube. Remove tourniquet. Draw a second green top tube. Invert gently 2-3 times. Discard the first tube. Separate plasma from cells in the second tube. Make sure tubes are silicone-coated glass. Traumatic venipuncture causing hemolysis is the cause for rejection of specimen. Centrifuge tube #2 at 1000 X g for 10 minutes. Draw off the supernatant plasma with a transfer pipet and deliver it into a second glass tube. Recentrifuge the plasma at 1600 X g for 20 minutes. Then carefully draw off the supernatant plasma and transfer it into a third glass tube. Label the tube. Keep refrigerated. If the assay is delayed, then freeze the sample below -20 oC. Deliver to Special Chemistry area, if possible."
