Preparation & Notes

Tissue requirements : Tissue preparation: The tissue should be formalin-fixed and embedded into a paraffin block using standard fixation methods with 10% neutral-buffered formalin. Other fixatives such as AZF, B5, Bouin’s, or Holland’s should not be used. Surface area: Tissue should have a surface area of at least 25 mm2 (5 x 5 mm2, 2.5 x 10 mm2). Tumor content: ≥ 20% If the ratio of nucleated malignant to nucleated non-malignant cells is too low, sensitivity of detection of certain variants and alterations is reduced . Additional Slide: Along with FFPE block, one additional slide stained with Hematoxylin and Eosin (H&E) should also be prepared marked with tumor content and surface area. Anwa OncoScope is a next generation sequencing (NGS) based test designed to empower personalized treatment decisions in patients with various solid tumors. It intended to detect targeted SNVs , INDELs, CNVs and rearrangement (gene fusions) in 524 genes (63 genes from NCCN/FDA cancer treatment guidelines, 116 cancer driver genes and 345 genes in vital cancer signaling pathways, as well as microsatellite instability (MSI) and tumor mutation burden (TMB). The detection is based on DNA and RNA, and utlizes unique molecular barcode (UMI) for digital error correction with limit of detection equal 1% allele frequency .
